Isolation and Characterization of a Thermophilic Oil-Degrading Bacterial Consortium

摘要 Inthisstudy,athermophilicoil-degradingbacterialconsortiumKO8-2growingwithinthetemperaturerangeof45-65℃(with55℃beingtheoptimumtemperature)wasisolatedfromoil-contaminatedsoilofKaramayinXinjiang,China.Denaturinggradientgelelectrophoresis(DGGE)showedthattherewereninestrainsincludedinKO8-2,whichoriginatedfromthegeneraofBacillus,GeobacillusandClostridium.Theyallbelongedtothermophilicbacteria,andhadbeenpreviouslyprovedasdegradersofatleastonepetroleumfraction.ThecrudeoildegradedbyKO8-2wasanalyzedbyinfraredspectrophotometry,hydrocarbongrouptypeanalysisandgaschromatography.TheresultsindicatedthatthebacterialconsortiumKO8-2wasabletoutilize64.33%ofsaturates,27.06%ofaromatics,13.24%ofresinsandtheoilremovalefficiencyreachedupto58.73%at55℃whentheoilconcentrationwas10g/L.DetailedanalysisshowedthatKO8-2wasabletoutilizethehydrocarboncomponentsbeforeC19,andthen-alkanesrangingfromC20-C33weresignifi-cantlydegraded.TheratiosofnC17/PrandnC18/Phwere3.12and3.87,respectively,beforedegradation,whereasafterdegradationtheratiosreducedto0.21and0.38,respectively.Comparedwiththecontrolsample,theoilremovalefficiencyinKO8-2compostingreactorreached50.12%afteradegradationdurationof60days.
