  • 简介:Twostarch-branchingenzyme(SBE)inrice,isknowntobeakeyenzymeinamylopectinbiosynthesis.ThecDNAoftwoSBE(starch-branchingenzyme)genesSheIandShedencodingSBEⅠandSBEⅢ(twomajorisoformsinrice)wereclonedbyanimprovedRT-PCRtechnique,fromatemplatecDNAlibray,derivedfromthetotalmRNAsextractedfromtheimmatureseedsofajaponicariceWuyunjing7.DNAsequenceanalysisshowedthatthesizeoftheclonedSheIandShedcDNAswere2490and2481bplong,respectively,includingtheirentirecodingsequences.ComparisonanalysisindicatedthatthenucleotidesequenceofShe3wasthesameasthatofshed(GenbankAccessionNo.D16201)asreportedpreviously.Therewereonlyfourbase-pairsdifference,whichresultedinchangesoftwodeducedaminoacidsbetweentheclonedShe1cDNAandthereportedshe1(GenbankAccessionNo.D11082).TheclonedSheIandShedcDNAsmakeitpossibletoimprovericestarchqualitythroughgeneticengineering.

  • 简介:Saltstressisamajorprobleminmostofthericegrowingareasintheworld.AmajorQTLSaltolassociatedwithsalttoleranceattheseedlingstagehasbeenmappedonchromosome1inrice.ThisstudyaimedtocharacterizethehaplotypediversityatSaltolandadditionalQTLsassociatedwithsalttolerance.Salttoleranceattheseedlingstagewasassessedin54ricegenotypesinthescaleof1to9scoreatEC=10dSm-1undercontrolledenvironmentalconditions.SevennewbreedinglinesincludingthreeKMR3/O.rufipogonintrogressionlinesshowedsimilarsalttolerantabilityasFL478andcanbegoodsourcesofnewgenes/allelesforsalttolerance.Simplesequencerepeat(SSR)markerRM289showedonlytwoallelesandRM8094showedsevenalleles.Polymorphicinformationcontentvaluevariedfrom0.55forRM289to0.99forRM8094andRM493.Basedon14SSRmarkers,the54lineswereclearlyseparatedintotwomajorclusters.FourteenhaplotypeswereidentifiedbasedonSaltollinkedmarkerswithFL478asthereference.AllelesofRM8094andRM3412candiscriminatebetweenthesalttolerantandsusceptiblegenotypesclearlyandhencecanbeusefulinmarker-assistedselectionattheseedlingstage.OthermarkersRM10720onchromosome1andRM149andRM264onchromosome8canalsodistinguishtolerantandsusceptiblelinesbutwithlesserstringency.

  • 简介:Thepromoterregionofadroughtandabscisicacid(ABA)induciblegene,osr40c1,wasisolatedfromasalt-tolerantindicaricevarietyPokkali,whichis670bpupstreamoftheputativetranslationstartcodon.Insilicopromoteranalysisofresultedsequenceshowedthatatleast15typesofputativemotifsweredistributedwithinthesequence,includingtwotypesofcommonpromoterelements,TATAandCAATboxes.Additionally,severalputativecis-acingregulatoryelementswhichmaybeinvolvedinregulationofosr40c1expressionunderdifferentconditionswerefoundinthe5′-upstreamregionofosr40c1.TheseareABA-responsiveelement,light-responsiveelements(ATCT-motif,BoxI,G-box,GT1-motif,Gap-boxandSp1),myeloblastosisoncogeneresponseelement(CCAAT-box),auxinresponsiveelement(TGA-element),gibberellin-responsiveelement(GARE-motif)andfungal-elicitorresponsiveelements(BoxEandBox-W1).Aputativeregulatoryelement,requiredforendosperm-specificpatternofgeneexpressiondesignatedasSkn-1motif,wasalsodetectedinthePokkaliosr40c1promoterregion.Inconclusion,thebioinformaticanalysisofosr40c1promoterregionisolatedfromindicaricevarietyPokkaliledtotheidentificationofseveralimportantstress-responsivecisactingregulatoryelements,andtherefore,theisolatedpromotersequencecouldbeemployedinricegenetictransformationtomediateexpressionofabioticstressinducedgenes.

  • 简介:Weedyriceexertsasevereimpactonriceproductionbycompetingforsunlight,waterandnutrients.Thisstudyassayedthepopulationstructure,geneticdiversityandoriginofNortheastAsiaweedyricebyusing48simplesequencerepeatmarkers.TheresultsshowedthatweedyriceinNortheastAsiahadahighgeneticdiversity,withShannon’sdiversityindex(I)of0.748andtheheterozygosity(He)of0.434.Ineachregionalpopulation,Ivaluevariedwidely.ThewidestrangeofI(0.228–0.489)wasobservedintheweedyriceofEasternChina,whichwaslargerthanthatofNortheastChinaandKorea(0.168–0.270).TheF-statisticsofregionalpopulations(Fis,FitandFst)alsoshowedhighervaluesintheweedyriceofEasternChinathanthoseofNortheastChinaandKorea.Allweedyriceaccessionsweregroupedintotwoclustersintheunweightedpairgroupmethodwitharithmeticmeanclusteranalysisdendrogram,namelyEasternChinabranchandNortheasternChinaplusKoreabranch.TherewassignificantdifferentiationingeneticcharacteristicsinweedyriceofnortheasternandeasternAsia,especiallyinEasternChina.

  • 简介:在中国在商业杂交稻生产推广的41根父母线的基因差异被使用词法特点和简单顺序重复(SSR)的簇分析学习标记。41个条目被分配进二簇(即早成熟ormedium簇;中等或迟了成熟的簇)并且进一步在词法特点簇分析上分配了sub-clustersbased进六。早或中等成熟的簇是镇静的of15维护者线,四根早成熟的恢复系线和二个thermo敏感的妖怪不育系,并且中等或迟了成熟的簇包括了成熟orlate维护者衬里的16根restorer线和4媒介。而且,SSR簇分析分类41条目intotwogroups(即维护者线组和恢复系线组)并且七亚群。维护者线组由所有19根维护者线组成了,二个thermo敏感的妖怪不育系,当restorer线组由所有构成时,20restorer排队。与家谱信息更好适合的SSR分析。从杂交稻繁殖上的看法,结果建议thatSSR分析可能是一个更好的方法在indica杂交稻学习父母线的差异。

  • 简介:Thelight-harvestingchlorophylla/b-proteincomplexplaysanimportantroleinphotosynthesisofplants.Afull-lengthcDNAoflight-harvestingchlorophylla/b(cab)genewasclonedfromthefirststrandofMoso(Phyllostachysedulis)cDNAthroughRT-PCRandRACEmethods,namedascab-PhE10(cabgene10fromPh.edulis).Thelengthofcab-PhE10(GenBankaccessionnumber:EU118754)is1151bp,whichcontainsanopenreadingframeencoding283aminoacidsfrom81stto932ndposition.Thebioinformaticsanalysisindicatedthattheproteinencodedbycab-PhE10hadachlorophlla/bbindingdomain(83rd-247thposition),twoproteinkinaseC-phosphorylationsites,threeN-myristoylationsitesandayiaA/Bdoublehelixdomain.Theaminoacidsequenceofcab-PhE10showedhighsimilaritywiththecabgenesofOryzasativa,Zeamays,Hordeumvulgare,andVitisvinifera,morethan80%,respectively,whichindicatedthatcab-PhE10genebelongstolhcb5genefamily.
