Granulometric Analysis of Spots in DNA Microarray Images

AsthetopologicalpropertiesofeachspotinDNAmicroarrayimagesmayvaryfromoneanother,weemployedgranulometriestounderstandtheshape-sizecontentcontributedduetoasignificantintensityvaluewithinaspot.Analysiswasperformedonthemicroarrayimagethatconsistedof240spotsbyusingconceptsfrommathematicalmorphology.Inordertofindoutindicesforeachspotandtofurtherclassifythem,weadoptedmorphologicalmultiscaleopenings,whichprovidedmicroarraysatmultiplescales.Successiveopenedmicroarraysweresubtractedtoidentifytheprotrusionsthatweresmallerthanthesizeofstructuringelement.Spot-wisedetails,intermsofprobabilityoftheseobservedprotrusions,werecomputedbyplacingaregularlyspacedgridonmicroarraysuchthateachspotwascenteredineachgrid.Basedontheprobabilityofsizedistributionfunctionsoftheseprotrusionsisolatedateachlevel,weestimatedthemeansizeandtextureindexforeachspot.Withthesecharacteristics,weclassifiedthespotsinamicroarrayimageintobrightanddullcategoriesthroughpatternspectrumandshape-sizecomplexitymeasures.Thesesegregatedspotscanbecomparedwiththoseofhybridizationlevels.
