  • 简介:Danshensu[3-(3,4-dihydroxyphenyl)lacticacid,DSS],oneofthesignificantcardioprotectivecomponents,isextractedfromtherootofSalviamiltiorrhiza.Inthepresentstudy,anesterprodrugofDanshensu(DSS),palmitoylDanshensu(PDSS),wassynthesizedwiththeaimtoimproveitsoralbioavailabilityandprolongitshalf-life.TheinvitroexperimentswerecarriedouttoevaluatethephysicochemicalpropertiesandstabilityofPDSS.AlthoughthesolubilityofPDSSinwaterwasonly0.055mg·mL~(-1),itssolubilityinFaSSIFandFeSSIFreached4.68and9.08mg·mL~(-1),respectively.Octanol-waterpartitioncoefficient(logP)wasincreasedfrom-2.48ofDSSto1.90ofPDSS.PDSSwasrelativelystableintheaqueoussolutioninpHrangefrom5.6to7.4.Furthermore,thepharmacokineticsinratswasevaluatedafteroraladministrationofPDSSandDSS.AUCandt1/2ofPDSSwereenhancedupto9.8-foldand2.2-fold,respectively,comparedtothatofDSS.Cmaxwas1.67±0.11μg·mL~(-1)forPDSSand0.81±0.06μg·mL-1forDSS.Thus,theseresultsdemonstratedthatPDSShadmuchhigheroralbioavailabilityandlongercirculationtimethanitsparentdrug.

  • 简介:Objective:TheaimofthisexperimentistostudytheeffectofthreemethodsofsurfacemodificationonthecorrosionresistanceofcommercialpureTitaniumwhenusedinoralenvironmentforhalfayear.Method:48specimensofpuretitaniumweremadeanddividedintofourgroupsrandomly,onegroupwasselectedrandomlyasGroupⅠ(controlgroup),theotherthreegroupsweretreatedbythreemethodsofsurfacemodificationindividually,GroupⅡ:heatingoxidationinair(400℃,30min.),GroupⅢ:anodization(45volts,10min.),GroupⅣ:TiNcoating(firingtemperature200℃,totalcoatingtime62min.).Sixedentulousvolunteerswithhealthyoralmucosaparticipatedintheinvivostudy.Onetestingpiecefromeachgroupwasselectedandfixedinthepolishedsurfaceofuppercompletedentures.Dynamicpolarizationcurvesweretracedwithelectrochemicalmethodafterthespecimenswereplacedeitherinoralcavityorinairfor6months.Results:Afterallspecimenswereused,Ecorralteredineverygroup,Ecorrfromhightolowwereinturn:TiNcoatinggroup>heatingoxidationgroup>anodizationgroup>controlgroup,noobviouspassivepotentialEpandIpwasfoundincontrolgroup.HeatingoxidationinairexhibitedsimilarEptoanodization,butIpwasremarkablylowerthanthatofanodization;TiNcoatingshowedobviouslydifferentpolarizationcurvescomparedwithheating-oxidationgroupandanodizationgroup,Ecorrwaspositive,andnoEpandIpwasfound.Conclusion:Underpresentexperimentalcondition,allthethreetreatmentmethodscouldenhancecorrosionresistanceofpuretitaniuminoralenvironment,heatingoxidationinairexhibitedbetterresistancetocorrodethananodization,TiNcoatingpossessedthemostexcellentcorrosionresistance,evenafterexposedinoralconditionfor6months,therewaslittlechangeofcorrosionresistance.ThereforeTiNcoatingcouldbeadoptedtoimprovecorrosionresistanceofpuretitaniuminoralenvironment.

  • 简介:AIM:Toinvestigatethedamagingeffectofhigh-intensityfocusedultrasound(HIFU)oncancercellsandtheinhibitoryeffectontumorgrowth.METHODS:MurineH22hepaticcancercellsweretreatedwithHIFUatthesameintensityfordifferentlengthsoftimeandatdifferentintensitiesforthesamelengthoftimeinvitro,thedeadcancercellsweredeterminedbytrypanbluestaining.TwogroupsofcancercellstreatedwithHIFUatthelowestandhighestintensitywereinoculatedintomice.Tumormasseswereremovedandweighedafter2wk,tumorgrowthineachgroupwasconfirmedpathologically.RESULTS:ThedeathrateofcancercellstreatedwithHIFUat1000W/cm2for0.5,1,2,4,8,and12swas3.11±1.21%,13.37±2.56%,38.84±3.68%,47.22±5.76%,87.55±7.32%,and94.33±8.11%,respectively.ApositiverelationshipbetweenthedeathratesofcancerceilsandthelengthofHIFUtreatmenttimewasfound(r=0.96,P<0.01).ThedeathrateofcancercellstreatedwithHIFUattheintensityof100,200,400,600,800,and1000W/cm2for8swas26.31±3.26%,31.00±3.87%,41.97±5.86%,72.23±8.12%,94.90±8.67%,and99.30±9.18%,respectively.ApositiverelationshipbetweenthedeathratesofcancercellsandtheintensitiesofHIFUtreatmentwasconfirmed(r=0.98,P<0.01).ThecancercellstreatedwithHIFUat1000W/cm2for8swereinoculatedintomiceexvivo.Thetumorinhibitoryratewas90.35%comparedtothecontrol(P<0.01).IntheexperimentalgroupinoculatedwiththecancercellstreatedwithHIFUat1000W/cm2for0.5s,thetumorinhibitoryratewas22.9%(P<0.01).Bypathologicalexamination,tumorgrowthwasconfirmedin8outof14mice(57.14%,8/14)inoculatedwiththecancercellstreatedwithHIFUat1000W/cm2for8s,whichwassignificantlylowerthanthatinthecontrol(100%,15/15,P<0.05).CONCLUSION:HIFUiseffectiveonkillingordamageofH22hepaticcancercellsinvitroandoninhibitingtumorgrowthinmiceexvivo.

  • 简介:AbstractObjective:To test the feasibility of a real time miniature endoscope system for imaging the nasopharynx.Study design:Preclinical assessment on skull model and cadaver.Methods:A 3.5 mm miniature endoscope was fabricated and the image capture of the nasopharynx was investigated by positioning the miniature camera system at the posterior free edge of the vomer bone. Wireless real time transmission of the images and quality was tested in a skull model. Next, three nasopharyngeal surveillance miniature camera system were developed for possible clinical translation. Two prototypes were anchored on the nasal septum and the last prototype was designed using a patient self-administered surveillance process. These prototypes were tested for feasibility on both the phantom skull and cadaveric model. Risk assessments were also performed to assess risk, safety and validate the reliability of the material utilized for clinical translation.Results:Insertion and anchorage of the miniature surveillance endoscope prototypes at the vomer bone were feasible on all 3 prototypes. The quality of captured images was reasonable and miniaturized camera was responsive to pan at different angles so that the entire nasopharynx may be surveyed. Risk assessments on the material such as pull out test, breaking force analysis, finite element test and tensile strength test were reliable for possible clinical translation.Conclusions:Real time miniature endoscope system for surveillance of nasopharyngeal cancer is feasible. Clinical translation of this technology was possible but requires further refinement in enhancing image quality and wireless transmission of the captured images.

  • 简介:AbstractWe analyzed variations in the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) genome during a flight-related cluster outbreak of coronavirus disease 2019 (COVID-19) in Shenzhen, China, to explore the characteristics of SARS-CoV-2 transmission and intra-host single nucleotide variations (iSNVs) in a confined space. Thirty-three patients with COVID-19 were sampled, and 14 were resampled 3-31 days later. All 47 nasopharyngeal swabs were deep-sequenced. iSNVs and similarities in the consensus genome sequence were analyzed. Three SARS-CoV-2 variants of concern, Delta (n = 31), Beta (n = 1), and C.1.2 (n = 1), were detected among the 33 patients. The viral genome sequences from 30 Delta-positive patients had similar SNVs; 14 of these patients provided two successive samples. Overall, the 47 sequenced genomes contained 164 iSNVs. Of the 14 paired (successive) samples, the second samples (T2) contained more iSNVs (median: 3; 95% confidence interval [95% CI]: 2.77-10.22) than did the first samples (T1; median: 2; 95% CI: 1.63-3.74; Wilcoxon test, P = 0.021). 38 iSNVs were detected in T1 samples, and only seven were also detectable in T2 samples. Notably, T2 samples from two of the 14 paired samples had additional mutations than the T1 samples. The iSNVs of the SARS-CoV-2 genome exhibited rapid dynamic changes during a flight-related cluster outbreak event. Intra-host diversity increased gradually with time, and new site mutations occurred in vivo without a population transmission bottleneck. Therefore, we could not determine the generational relationship from the mutation site changes alone.

  • 简介:AbstractBackground:Angiogenesis and hypoxia-inducible factor 1α (HIF-1α) play major roles in solid tumors. This study aimed to establish a longitudinal and multimodal imaging model for in vivo evaluation of HIF1α and angiogenesis in breast cancer.Methods:By transfection of a 5 hypoxia-responsive element (HRE)/green fluorescent protein (GFP) plasmid, the cell line Ca761-hregfp was established, which emitted green fluorescence triggered by HIF-1α under hypoxia. The cells were subjected to CoCl2-simulated hypoxia to confirm the imaging strategy. We grew Ca761-hre-gfp cells in the left rear flanks of twelve 615 mice. Experiments were conducted on days 4, 9, 15, and 19. For in vivo analysis, Ca761-hre-gfp subcutaneous allografted tumors were imaged in vivo using contrast-enhanced ultrasound (CEUS) and fluorescence imaging (FLI) during tumor development. The tumor size, CEUS peak intensity, and FLI photons were measured to evaluate tumor growth, angiogenesis, and HIF-1α activity, respectively. After each experiment, three mice were randomly sacrificed and tumor specimens were collected to examine HIF-1α activity and the microvessel density (MVD).Results:In vitro, both green fluorescence and HIF-1α expression were detected in Ca761-hre-gfp cells treated with CoCl2, indicating the suitability of the cells to detect HIF-1α activity. In vivo, HIF-1α activity first increased and then decreased, which was significantly correlated with angiogenic changes (r = 0.803, P = 0.005). These changes were confirmed by immunohistochemical staining of HIF-1α and MVD.Conclusions:The findings validated the Ca761-hre-gfp murine allograft model for reliable evaluation of HIF-1α activity and angiogenesis longitudinally using both molecular and pre-clinical non-invasive imaging modalities. The cell line may be useful for studies of anti-HIF pathway therapies.

  • 简介:Theanticoagulanteffectofleechsalivawastraditionallyemployedinthetreatmentofdiabetesmellituscomplicationssuchasperipheralvascularcomplications.Thisstudywascarriedouttoexaminetheeffectofleechsalivaextract(LSE)onbloodglucoselevelsinalloxan-induceddiabeticrats.First,LSEwascollectedfromleecheswhichwerefedonaphagostimulatorysolution.Second,totalproteinconcentrationwasestimatedusingtheBradfordassay.Third,diabeticratswereinjectedsubcutaneously(sc)withLSEatdosesof500and1000μg·kg1bodyweight(bw).Otherdiabeticratswereinjectedscwithinsulinatdosesof10and20U·kg1bw.AnothergroupwasinjectedsimultaneouslywithLSE(250μg·kg1bw)andinsulin(10U·kg1bw).Fastingbloodglucose(FBG)concentrationsweremonitoredduringastudyperiodofeighthoursatregularintervals.FindingsshowedthatbothdosesofLSEresultedinasignificantandgradualdecreaseinFBGstartingfrom10%18%downfallaftertwohoursofinjectionreachingthemaximalreductionactivityof58%aftereighthours.Remarkably,LSEwassufficienttobringtheratstoanearnorm-glycemicstate.Thehighdoseofinsulininducedaseverehypoglycemicconditionafter24hofinjection.ThelowerdosewasabletodeclineFBGfor26hinratswhichbecamediabeticagainafter8h.Ontheotherhand,theconcurrentinjectionoflowdosesofLSEandinsulinproducedahypoglycemiceffectwithallratsshowingnormalFBGlevels.Takentogether,thesefindingsindicatedthatthesubcutaneousinjectionofLSEofthemedicinalMalaysianleechwasabletoprovidebetterglycemiccontrolcomparedwithinsulin.Moreover,thesynergismbetweenLSEandinsulinsuggeststhatLSEcouldbeutilizedasanadjuvantmedicationinordertoreduceinsulindosageortoachievebettercontrolofbloodglucose.
